Limitations of Traditional Scale-Up
This is a significant departure from what has, historically, been standard practice in preparative chromatography. Traditionally, separations were developed on an analytical column packed with a small particle spherical phase and transferred to a preparative column packed with a larger particle irregular phase.
The analytical and preparative phases often differed in pore size and carbon load. These differences might cause the selectivity and retention characteristics of these columns to differ dramatically. Often methods developed on the analytical column had to be redeveloped or significantly modified in order to provide adequate separation on the preparative column.

R&D Sets Eliminate Scale-Up Problems
The use of YMC R&D column sets eliminates potential selectivity differences caused by different types of silica and different particle and pore size packings by providing matched columns packed from the exact same lot of packing material. For example, if the preparative column is to be packed with a 15 micron spherical ODS phase, YMC will provide a matched analytical column packed with the same lot of 15 micron spherical ODS.

Ordering Information
R&D scale-up kits always consist of two columns, one analytical column (4.6 mm i.d.) and one preparative column. Each R&D scale-up kit has an individual “column dimension code” (please refer to the first columns of the two charts on the left), which describes its combination of column dimensions. In these two charts, the commonly used R&D scale-up kit column combinations are listed. Other combinations are available on request.
